Inspiré par des faits réels, Afronauts raconte une histoire alternative de la course vers l'espace dans les années 1960. La nuit du 16 juillet 1969, alors que l'Amérique se prépare à envoyer Apollo 11 sur la lune, un groupe d'exilés dans le désert zambien font leur possible pour lancer leur fusée les premiers. Matha Mwamba, 17 ans, est toute désignée. Elle devra décider d'accepter ou pas le prix à payer pour entrer dans la légende.
